The six-episode series will pick up three decades after the original 1995 film, following Cher as a successful businesswoman and mother whose daughter's teenage years leave her feeling "clueless" all over again.
Filming is expected to begin in Los Angeles in 2027.
The project was originally in development at Peacock before moving to Paramount+, the home of the original film.
Silverstone will also serve as an executive producer alongside Clueless writer and director Amy Heckerling and producer Robert Lawrence.
The new series is being written by Josh Schwartz, Stephanie Savage and Jordan Weiss, with Paramount+ describing the production as a homecoming for one of its most beloved franchises.
No additional cast members have been announced.
Released in 1995, Clueless became a defining teen comedy of the decade, loosely inspired by Jane Austen's Emma.
The film also launched the careers of stars including Paul Rudd, Brittany Murphy and Stacey Dash, and has remained a pop culture favourite ever since.
