Speaking at the London premiere of Spider-Man: Brand New Day, the actress described Holland as her "best friend" and said sharing the franchise with him had been "a privilege".
Asked what it's like to work with her husband, Zendaya replied: "When you're best friends, it's pretty easy."
The pair first met after being cast in Spider-Man: Homecoming in 2016 and have since starred together in several Marvel films.
Their relationship became public in 2021, and they have recently reunited on screen in both The Odyssey and the newest Spider-Man instalment.
Holland, who has portrayed Peter Parker for almost a decade, said returning to the role remains exciting because each film brings something new.
While he declined to say whether Brand New Day would mark the end of his time as Spider-Man, he said he still hopes to share the screen with more Marvel characters in future projects.
The latest film also introduces Stranger Things star Sadie Sink in a mystery role that has sparked widespread speculation among fans.
The story follows the events of the previous Spider-Man film, in which Peter Parker erased himself from the memories of those closest to him.
